The incidence of inflammatory bowel diseases (IBD) has risen dramatically in recent decades. Moreover, the condition is increasingly affecting young children, many of which present with an aggressive and treatment resistent phenotype. Environmental factors are thought to play a key role in disease pathogenesis and hence, epigenetic mechanisms are increasingly being proposed as the molecular mechanisms capable of mediating external triggers into stable phenotypic changes. Our group is particularly interested in the role of DNA methylation in regulating intestinal epithelial cell function during health and paediatric IBD. In addition to performing genome wide DNA methylation profiling of purified primary human intestinal epithelium from children diagnosed with IBD, we are in the process of establishing human 3D intestinal epithelial organoids as a cell type and disease specific model to investigate IBD pathogenesis.
